ABSTRACT

The current document is based on a consensus reached by a panel of experts from the Chinese Society of Allergy and the Chinese Society of Otorhinolaryngology-Head and Neck Surgery, Rhinology Group. Chronic rhinosinusitis (CRS) affects approximately 8% of Chinese adults. The inflammatory and remodeling mechanisms of CRS in the Chinese population differ from those observed in the populations of European descent. Recently, precision medicine has been used to treat inflammation by targeting key biomarkers that are involved in the process. However, there are no CRS guidelines or a consensus available from China that can be shared with the international academia. The guidelines presented in this paper cover the epidemiology, economic burden, genetics and epigenetics, mechanisms, phenotypes and endotypes, diagnosis and differential diagnosis, management, and the current status of CRS in China. These guidelines—with a focus on China—will improve the abilities of clinical and medical staff during the treatment of CRS. Additionally, they will help international agencies in improving the verification of CRS endotypes, mapping of eosinophilic shifts, the identification of suitable biomarkers for endotyping, and predicting responses to therapies. In conclusion, these guidelines will help select therapies, such as pharmacotherapy, surgical approaches and innovative biotherapeutics, which are tailored to each of the individual CRS endotypes.

ABSTRACT

Objective:To investigate whether diethyldithiocarbamate (DDC) affect the activity of superoxide dismutase (SOD) of Bifidobacterium infantis ( BB ) .Methods: We used simple anaerobic activated ways to culture BB .After the bacterium reached platform growth period ,two concentrations (200 and 1 000 μmol/L) of DDC were added to bacterium medium for a specific period of time (0.5,1,2,4 and 24 h),which was in order to determine if this inhibitor affected the activity of SOD .As a positive control,the in-hibitory effect of DDC on the activity of SOD in murine intestinal samples was also checked .Results:It was found that incubation of BB with DDC for some specific periods showed no effect on the activity of SOD .However , the activity of SOD in intestine reduced dramatically after the mice were given DDC in vivo .Conclusion:DDC affect the SOD activity in mice ,while it showed no effect on SOD activity of BB.The suitable inhibitor for SOD of BB is not commercially available and further research is warranted .

ABSTRACT

Objective: To clone , express and purify of Dermatophagoides farinae ( Der f 11 ) , and then test its immunogenicity.Methods:The gene coding for Der f 11 was synthesized ,and was then linked with the pET-32a vector.The expression plasmid pET32a(+)-Der f 11 was induced by IPTG.After purification of recombinant allergens Der 11 proteins through the Ni +affinity chromatography ,immunological allergic patients serum as the Primary antibody.Results: We obtain high purity recombinant Der f 11 protein.The results of SDS-PAGE show that the expression product is about 118 KD.Recombinant allergen Der f 11 test 15 dust mites allergic patients serum specific IgE , positive rate was 20%.Conclusion: Recombinant allergen Der f 11 obtained has the similar immunologic activity to natural Der f 11 protein.It can lay the foundation for the specific diagnosis ,treatment and further experimental studies of the dust mite allergy disease.

ABSTRACT

Objective:To isolate,identify and purify the Artemisia sieversiana pollen ,the mostly widespread pollen among the Artemisia pollens in China.Methods: Artemisia sieversiana extract was precipitated by saturated ammonium sulfate and then electrophoresed by SDS-PAGE.The molecular mass of each protein band was determined by gel media system.The primary allergen proteins were identified by Western blot.Allergen proteins were purified and identified by DEAE-cellulose DE-52 ion exchange chroma-tography ( IEC) and Western blot.Results: We isolated more than twenty protein bands from Artemisia sieversiana pollen extract , including the most abundant six bands whose Mr were 62 kD,57 kD,38 kD,29 kD,25 kD,14 kD espectively.The protein bands with Mr were 62 kD and 16 kD had the highest binding capacity with the specific IgE from Artemisia pollen allergic patients.The DEAE-cellulose DE-32 IEC was used to purify the primary allergen proteins with Mr 62 kD and 16 kD.Conclusion:The primary allergens of Artemisia sieversiana include the allergen proteins whose Mr are 62 kD,16 kD and the allergen of Mr 62 kD and 16 kD can be purified by chromatography.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the role of TIM4 (T cell immunoglobulin and mucin domain molecule 4) in the pathogenesis of allergic rhinitis (AR) in mice, and to identify a novel therapeutic target for the treatment of AR.</p><p><b>METHODS</b>Twenty-one male BALB/C mice of clean grade were divided into three groups randomly (n = 7 per group) including control, AR and anti-TIM4 antibody treatment groups. In order to induce upper airway allergic inflammation, the mice from AR and anti-TIM4 antibody treatment groups were sensitized by intraperitoneal injection followed by intranasal challenge with ovalbumin. Before the ovalbumin challenge, a group of mice was treated with anti-TIM4 antibody. To assess the AR model, behavioral observation with immunological assessments and HE staining of nasal tissues were performed. The TIM4 expression in nasal tissues in different groups of mice were assessed by immunofluorescence and RT-PCR.SPSS18.0 software was used to analyze the data.</p><p><b>RESULTS</b>The AR model in mice was successfully established as shown by behavioral observation and immunological evaluation. RT-PCR assays showed the relative expression of TIM4 mRNA in nasal mucosa of AR, control and anti-TIM4 antibody treatment mice was 16.29 ± 3.80, 0.51 ± 0.60, 1.64 ± 0.98, respectively. There was statistically significant differences mong three group (F = 46.56, P < 0.05). The expression of TIM4 in AR group was significantly higher than those in control group (t = 8.650, P < 0.05) and anti-TIM4 group (t = 8.027, P < 0.05). The expression of TIM4 was significantly reduced in the anti-TIM4 antibody group, as well as control group (t = -0.623, P > 0.05). More expression of TIM4 was detected in local nasal tissues of AR mice, mainly located below the pseudostratified ciliated columnar epithelium.</p><p><b>CONCLUSIONS</b>TIM4 plays a crucial role in the pathogenesis of AR. Effective inhibition of TIM4 expression can partially reverse the pathological changes of AR.</p>

ABSTRACT

In order to gain insight into a possible association between chronic sinusitis and asthma,85 patients with sinusitis and asthma underwent functional endoscopic sinus surgical treatment and serum antibodies and cytokines were measured. The results showed that 51 out of 85 patients with high serum anti-Staphylococcus enterotoxin B (SEB) antibody before treatment obtained satisfactory results for both sinusitis and asthma. The high level of Th2 cytokine IL-4 was down regulated tothe levels of normal controls after sinus surgery. Thirty-four out of 85 patients did not show high serum anti-SEB antibody before sinus surgery and did not show much improvement in their asthmatic symptoms although sinusitis symptoms were resolved by sinus surgery. It was concluded that bacterial superantigen SEB (in the sinuses) might play a crucial role in the pathogenesis of lower airway hypersensitivity.
